چکیده

Inhibition of immune precipitation with oligosaccharides obtained from Candida stellatoidea mannan has been used to provide more information about the haptenic groups of this serologically active polysaccharide. The oligosaccharides di-, tri-, tetra-, and a mixture of penta- and hexasaccharides were studied. The inhibitory activity of these materials was tested with two immune sera. With one serum, 0.8 mumole of the mixture of penta- and hexasaccharides inhibited the reaction by 87%, and, with the other serum, 0.4 mumole of the mixture inhibited the reaction by 99%. Monosaccharides were also tested with each antiserum and found to be noninhibitory. It is apparent that the mixture of oligosaccharides containing 5 to 6 mannose units was the most effective inhibitor. Since it is known that these oligosaccharides contain a predominance of alpha 1-2 linkages and lesser numbers of alpha 1-3 linkages, it is likely that these are important in the structural configuration of the antigenic determinants.

Immunosorbent for the isolation of specific antibodies against mannan: localization of antigens in yeast cell walls.

A procedure is given for the insolubilization of yeast mannan by entrapment in a polyacrylamide gel. Specific anti-mannan antibodies were separated by immunosorption from all other antibodies of a rabbit antiserum against whole, Formalininactivated Candida utilis cells. Colloidal gold coated with the anti-mannan and anti-non-mannan antibody fractions was used for the ultrastructural visualizati...

Isolation of Mannooligosaccharides Corresponding to Antigenic Determinants of Pathogenic Yeast Candida catenulata Cell Wall Mannan

To investigate the chemical structure of cell wall mannan of pathogenic yeast, Candida catenulata IFO 0745 strain, which possess the epitopes of antigenic factors 1, 9, and 34 to genus Candida, we previously performed the two-dimensional nuclear magnetic resonance (NMR) analysis of this mannan, Fr. C, without the need for harsh procedures. In this study, three oligosaccharides, biose, triose, a...

Growth inhibition of Candida albicans in the presence of antiserum elicited in rabbits by mannan-protein conjugate.

Antifungal properties of rabbit antiserum prepared by immunization are reported. The immunization was done by a chemically prepared conjugate consisting of Candida albicans (serotype A) surface mannan and human serum albumin. Addition of rabbit antiserum to D-glucose medium inoculated with C. albicans effectively inhibited its growth. Moreover, C. albicans cells treated with rabbit antiserum re...

Identification of two Saccharomyces cerevisiae cell wall mannan chemotypes.

We have obtained evidence for two structurally and antigenically different Saccharomyces cerevisiae cell wall mannans. One, which occurs widely and is found in S. cerevisiae strain 238C, is already known to be a neutral mannan which yields mannose, mannobiose, mannotriose, and mannotetraose on acetolysis of the (1 --> 6)-linked backbone. The other, which was found in S. cerevisiae brewer's stra...

Relationship between phosphate content and immunochemical properties of subfractions of bakers' yeast mannan.

The mannan of bakers' yeast (Saccharomyces cerevisiae) was fractionated on a column of diethylaminoethyl-Sephadex into five subfractions. Phosphate content of these mannan subfractions was proportional to the concentration of NaCl solutions used in the chromatographic separation. Quantitative precipitin reactions showed that the serological reactivities of the subfractions were proportional to ...
